摘 要:
双向启动子因为可以同时启动其两侧基因的表达而受到人们越来越多的关注。基因组结构分析及生物信息学研究发现拟南芥At3g21370与At3g21380基因共享启动子区, 是一个种子特异性双向基因对。序列分析发现这两个基因的间隔序列即共享启动子区含有GCN4和Skn-1等多个标志性种子特异性调控元件。将该间隔序列正反向同时与GFP和GUS报告基因融合构建双向报告基因表达载体。GUS组织化学染色和GFP荧光检测分析结果表明该间隔序列能够驱动外源双向基因对特异性共表达于种子中, 是一个种子特异性双向启动子。关键词:双向启动子; 顺式作用元件; 组织特异性; 种子

Abstract:
Bidirectional promoters have received more attention because of their ability to express simultaneously two downstream genes. Genome structure analysis and microarray-based studies indicated that the Arabidopsis genes At3g21370 and At3g21380 are a seed-specific bidirectional gene pair, which share a promoter region. Sequence analysis revealed that the shared promoter region of these two genes contains various seed-specific regulatory elements including the GCN4 and Skn-1 elements. Using a GFP:Promoter:GUS transgene approach has revealed that the intergenic region of bidirectional genes At3g21370/At3g21380 is a seed-specific bidirectional promoter, which could simultaneously drive GFP and GUS expression in the seeds of the transgenic Arabidopsis plants.Key words: bidirectional promoter; cis acting element; tissue-specific; seed
